Lipase Entrapped in Acrylamide Gel Lattice for Hydrolysis af Coconut Oil

This paper discusses the search for an adsorbent to be used as support material for a crude lipase preparation. Entrapment of the lipase into acrylamide gel lattice gave an enzyme preparation with high activity and amount of immobilized enzyme protein. The properties of the immobilized lipase were investigated and compared with those of the crude lipase. The pH and temperature optima shifted from 6.0 to 5.0 and 40°C to 35°C, respectively upon immobilization. The kinetic study of the immobilized lipase showed a higher in Km value, but the same Vmax compared with crude lipase. The enzyme was still active after the sixth reaction cycle using a shake flask system, indicating the enzyme stability towards repeated use.
Lipolytic action of the free and immobilized lipase on coconut oil was monitored by gas-liquid chromatography. An increase of 12% on the degree of hydrolysis was noted when immobilized lipase was used.
